Hypoxia-inducible Factors HIF-1 and HIF-2 in Head and Neck Cancer: Relationship to Tumor Biology and Treatment Outcome in Surgically Resected Patients
نویسندگان
چکیده
Hypoxia within head and neck squamous cell carcinoma (HNSCC) predicts a poor response to radiotherapy and poor prognosis. Hypoxiainducible factor (HIF)-1 and HIF-2 are nuclear transcription factors that regulate the cellular response to hypoxia and are important for solid tumor growth and survival. Overexpression of HIF-1 and HIF-2 was demonstrated in three HNSCC cell lines under hypoxia and tumor tissue versus normal tissue (n 20, HIF-1 , P 0.023; HIF-2 , P 0.013). On immunostaining, HIF-1 and HIF-2 expression were localized to tumor nuclei; HIF-2 expression was also seen in tumor-associated macrophages. Expression of HIF-1 in surgically treated patients with HNSCC (n 79) was associated with improved disease-free survival (P 0.016) and overall survival (P 0.027).

Blockade of Hypoxia: The Impact on Tumor Growth in an Experimental Tumor Model
Background: Tumor microenvironment is an active factor participating in immunoregulation, thereby preventing immunosurveillance and limiting the efficacy of anticancer therapies. Hypoxia as a major characteristic of solid tumors causes the expression of Hypoxia-Inducible Factor-1α (HIF-1α). This is a transcription factor that mediates hypoxic responses of tumor cells and involves in the express...
متن کاملThe transcription of Hypoxia-inducible factors (hif-1, hif-2) during normal development of Beluga sturgeon (Huso huso Linnaeus, 1758)
In the present study, the levels of the hypoxia-inducible factors (hif-1, hif-2) transcripts were evaluated during development of an ancient fish species, Beluga sturgeon (Huso huso) in a normal oxygen environment. The transcripts of hif-1, hif-2 were detected in all developmental time-points of H. huso from embryos to juvenile fish. Changes in normalized hif-1, hif-2 mRNA expression by using t...

Angiogenesis, as the process of new vessel formation from pre-existing vessels is dependent on a delicate equilibrium between endogenous angiogenic and antiangiogenic factors. However, under pathological conditions, this tight regulation becomes lost which can result in the formation of the different diseases such as cancer. Angiogenesis is a complex process that includes many gene products tha...

In this study, the effects of sublethal concentrations of the Endosulfan on gene expression of hypoxia-inducible factor (HIF-1 α, HIF-2 α) and vascular endothelial growth factor (VEGF) in the gill tissue of Persian sturgeon fingerling (Acipenser persicus) were studied. A total of 360 sturgeons (average weight of 2 ± 0.12 g) exposed to concentrations of 10, 20 and 40 micrograms per liter and VEG...
متن کاملInteraction between hypoxia, AKT and HIF-1 signaling in HNSCC and NSCLC: implications for future treatment strategies
BACKGROUND Hypoxia is a negative prognostic factor and this study investigated the relationship between hypoxia, hypoxia inducible factor 1 (HIF-1) and AKT signaling in head and neck squamous cell carcinoma (HNSCC) and non-small-cell lung cancer (NSCLC). RESULTS/METHODOLOGY pAKT was induced by hypoxia (0.5% O2) in a part of HNSCC (3/4) and squamous (2/3) and adenocarcinoma (1/3) NSCLS lines. ...
